Homocysteine is formed when methionine is metabolized to cysteine. Homocysteine metabolism is explained in the steps given below:

  • Methionine reacts with ATP and forms  S-Adenosyl-L-methionine (SAM). this is the universal methyl donor. After donating the methyl group to acceptor molecules, S-Adenosyl homocysteine (SAH) is formed which undergoes deadenosylation and forms homocysteine.
  • Remethylation pathway: The Homocysteine formed is remethylated back to Methionine utilizing the methyl group from 5-N-methyl tetrahydrofolate. This pathway requires folate, vitamin B12, and betaine.
  • Transsulfuration pathway: This process requires vitamin B6. Homocysteine combines with serine to form cystathionine.
